- STUDIES ON DECOMPOSING PHENOL BY IMMOBILIZED CELLS OF CANDIDA TROPICALIS No. 314
- 摘要:1.比较了三种生长培养基对热带假丝酵母314号细胞分解苯酚活力的影响。在以苯酚为碳源的培养基中生长的细胞酶活力较高,125mg湿细胞一小时可分解苯酚5425.8μg。用3.0%的琼脂固定化后,固定化细胞酶的相对活力为44.4%。诱导能显著地提高固定化细胞的酶活力,最适的诱导时间为12—18个小时。 2.比较了固定化细胞和自然细胞的某些性质。两种细胞反应的最适温度均为37℃;反应的pH范围均在3.5—11之间;固定化细胞热稳定性稍差;Cu2+和Zn2+对自然细胞的活性有一定抑制作用;CN-明显抑制两种细胞酶活力。 3.固定化细胞装柱后连续处理浓度约为150ppm的苯酚溶液,20天内的出水酚浓度平均为0.29ppm,在此条件下固定化细胞的半衰期为11天。 4.连续处理苯酚过程中,固定化细胞所丧失的酶活力可以通过再培养得到恢复。
- Abstract:1. Three different kinds of media on phenol-decomposing activity by cells of Candida tropicalis No. 314 were investigated. Activity of cells which grow in medium containing phenol as carbon source is high, 1 gram of the wet cells being able to decompose 43.4mg of phenol/hour. Relative activity of immobilized cells entrapped in an agar gel is 44.4%. The enzymatic activity of immobilized cells can be markedly increased by induction.optimum time for which is 12-18 hours.2. Some properties of immobilized cells were compared with those of native cells. Optimum temperature for enzymatic reaction is 37℃ for both cells. And optimum pH lies in the range of 4-11 for native cells, while 5-11 for immobilized cells. Inhibitory action of Cu2+and Zn2+(1×10-8M)on activity of native cells was shown to a certain extent. Activity of both immobilized and native cells was evidently inhibited by cyanide.3. The immobilized cells column has been used for continuous treatment of phenol(150 ppm)for 20 days at 30℃. Average concentration of phenol in effluent is 0.29 ppm with a half life of 11 days.4. The enzymatic activity of immobilized cells which has been lost in continuous treatment of phenol can be regenerated by reincubation.
