  • 摘要:本文是用染色体畸变和姊妹染色单体交换(SCE)的方法,检测了广谱抗肿瘤新药,平阳霉素(PYM),诱发哺乳动物细胞染色体畸变的能力,以便确定它的诱变性。 结果表明,PYM显著延长细胞分裂周期,大大提高染色体畸变频率,明显增加SCE的频率。显然,它对动物细胞具有强烈的诱变效果。
  • Abstract:Pingyangmycin (PYM) as a new carcinoma-curing drug may produce tremendous influence on chromosomal aberrations in plant cells, It therefore seems highly necessary to assess further whether this substance shows mutagenic and/or carcinogenic risk to human health.Exponential-growing Chinese Hamster ovary cells (CHO-KI) were thus treated with PYM (0.4 and 1.0μg/ml)in vitro for 40 hr in presence of BrdU (bromodexyuridine 7.0 /μg/ml) in Eagle's medium, and control was CHOKI without PYM treatment.During last 4hr of the treatment, colcemid (0.4 μg/m1 was added. PYM-treated cells were centrifuged, and stained with Giemsa on slides. The 1st and 2nd metaphases could be clearly distinguished because of bifilarly BrdU-substituted Chromatids showing a paler stain with Giemsa. Cultures were all treated with PYM and BrdU, and then sister chromatid exchanges (SCE) and chromosomal aberrations were noticed on the same preparations.Cell division was apparently inhibited by PYM. With the metaphase cell number of the 1st division increasing in proportion to PYM concentration, and by exposing to PYM at a conc, of 1.0μg/ml, chromosomal aberrations were increased about 4 folds as much as the control. The principal aberrations observed were single chromatid gaps, deletions, interchanges and chromosome rings, breakages, especially chromosome disintegrations, the frequencies of which have increased proportionally with concentrations of PYM.Frequency of SCEs was approximately doubled after exposure to PYM compared with the control.
