  • 摘要:本文比较了氯化镉对两种去分化和五种分化的鼠肝肿瘤细胞株的细胞毒性以及对金属硫蛋白的诱导能力。结果表明,经2μM氯化镉处理18h后,90—95%的细胞浆中镉同金属硫蛋白结合,而只有5—10%的镉同高分子量部分相连。在IIE细胞中镉与金属硫蛋白的结合取决于氯化镉的浓度与处理时间。当氯化镉浓度高达20μM时对去分化细胞的存活率未见有明显影响,但当浓度为3—10μM时则对分化细胞的存活率有显著影响。这种细胞毒性似与高分子量部分结合的镉的量有关,且氯化镉对这两种细胞的金属硫蛋白有不同的诱导能力。
  • Abstract:Five differentiated and two dedifferentiated rat hepatoma cell lines were compared in their capacities to induce cadmium-binding to metallo-thionein (MT) and high molecular weight protein.18 hours after incubation with 2μM CdCl2, the major portion of cytosolic Cd (90-95%) was associated with MT whereas only 5-10% of cytosolic Cd was bound to high molecular weight portion. The Cd-binding in MT fraction in HE cells was dependent on the time of exposure and concentration of CdCl2. The two types of cell lines varied in their sensitivities to Cd toxicity. Cloning efficiency of the dedifferentiated cell lines was not influenced up to about 20μM CdCl2 while the cloning efficiency of the differentiated cell lines decreased at 3-10μM CdCl2. The amount of HMW bound Cd seems to be correlated with Cd cytotoxicity and difference in inducibility of Cd-binding protein has been observed between the two types of rat hepatoma cell lines.
