假单胞杆菌抗镉基因的克隆

- 摘要:以pBR322为载体,将抗镉的假单胞杆菌R4染色体的抗镉基因克隆至大肠杆菌HB101,从5000个重组体中筛选出一株抗镉基因克隆株,命名为大肠杆菌HB101(pBC311)。重组质粒pBC311经PstⅠ酶解和琼脂糖凝胶电泳证实抗镉基因位于3.6kb长的PstⅠ片段上。克隆菌株可在含100μg/ml CdCl2的L-肉汤中生长,其生长停滞期比受体大肠杆菌HB101短得多,说明克隆菌株含有抗镉基因。
- Abstract:The Cd-resistant gene from Pseudomonas sp.R4 chromosome has been cloned using Pst I digested pBR322 as vector.The cloned gene is located on a 3.6kb fragment of the chromosome DNA according to an analysis on the restriction fragments of the recombinant plasmid pBC311.The cloned strain E.Coli HB101,pBC311grew well in L-broth containing 100 μg/ml CdCl2 with a much shorter lag phase than the parental strain E.Coli HB101 did.A restriction map of the cloned Cd-resistant gene was built, which shows a unique site for EcoRI, two sites for BamHI and Hindi and three sites for HinfI.
