- 摘要:DNA加合物是具有亲电性化学致癌物与DNA形成共价相联的化合物,是化学致癌物损伤DNA的主要形式,能够灵敏准确地分析DNA加合物的方法是研究DNA损伤.
- Abstract:Reaction of DNA-styrene oxide produced six adducts as determined by P1-nuclease mediatedi enhancemet of sensitivity of 32P-post-labeling in vitro. In this procedure, nuclease P1 cleaves deoxyribo-nucleoside 3'-monophosphates of normal nucleotides to deoxy-ribo-nucleosides which do not serve as substrate for polynucleotide ki-nase,while most adduct nucleotides were found to be totally or partially resistant to the 3'-dephosphorylating action of nuclease P1. Five adducts were detected quantitatively with a 150 to 800 fold increase in 32P-count rates as compared with the standard procedure (without P1 nuclease).
