  • 摘要:光合细菌球形红假单胞菌(Rhodopseudomonassphaeroides)和酿酒酵母(Saccharomycescerevisiae)细胞分别经溶茵酶和蜗牛酶脱壁形成原生质体,在聚乙二醇(PEG)和Ca2+诱导下实现了真核与原核细胞的超远缘杂交。经抗菌素培养基初步鉴定,融合产物能同时在含有链霉素和制霉茵素的培养基中生长,而双亲细胞只能在含有单一抗菌素的培养基中生长,经初筛与复筛后,获得高絮凝性融合(杂合)子菌株F13-1,F15-1和F20-1,将它们分别引入味精废水进行发酵反应,测定菌体转化有机废物生成生物量的效率即观察产率系数Yobs;测定单位菌体单位时间内在BOD5为3756mg/L味精废水中的菌体比增长率和有机污染物的比降解率。结果表明,3株杂种细胞在高浓度废水中能呈现出较高的合成生物量活性,而降解去除BOD5的性能不及双亲或介于双亲之间。
  • Abstract:The protoplast of photosynthetic bacteria (Rhodopseudomonas sphaeroids) and yeast (Saccharomyces cerevisiae) were obtained by using lysozyme and snail enzyme to degrade the cell wall respectively. The protoplasts belong to prokaryote and eukaryote, two types of kingdoms microorganisms. They were fused induced by polyethyleneglycol (PEG, MW = 6000) and CaCl2. the fusants could grow on the media which contained both nystatin (100 μ/mL) and streptomycin (100 μ/mL).But the parental strains could only grow on the media with one of the antibiotics. High flocculation hybrid cells F13-1, F15-1 and F20-1 were introduced into the fermentation system of monosodium glutamate (MSG) wastewater after the first and second screening. The observation growth yield (Yobs),specific growth rate (μ) and specific degradation rate (q) were measured in the wastewater with BOD5of 3756 mg/L. The results showed that the μ value of hybrid cells in the MSG wastewater was higher than that of their parental strains, but the q values of them are either lower than or similar to those of their parental strains.
