用苯及其生物代谢物酚、醌处理大鼠各器官中DNA加合物的形成和分布
- FORMATION AND DISTRIBUTION OF DNA ADDUCT IN ORGANS OF RAT TREATED WITH BENZENE,PHENOL, OR BENZOQUINONE
- 摘要:用苯及其在生物体内代谢物酚,醌处理大鼠,24h后取其肝、肾、肺、脾诸组织,用32P后标记方法测4种器官中DNA加合物含量,其结果是:在上述各器官中均发现有DNA加合物的形成,4种器官中总加合物量值对苯、酚、醌依次为9.18-89.13、20.9-243.8、53.1-308.9加合物/108正常核酸,3种系列化合物对大鼠4种器官中DNA加合物形成能力依次为醌>酚>苯。而大鼠中4种器官对同一化合物的加合物形成能力依次为肝>肾>肺>脾。这种结果较好地反映出3种系列化合物的代谢过程和生态毒理效应的差异性。
- Abstract:DNA adducts produced in various organs (liver, kidney, lung, and spleen) of SD rats induced either by Areolar and administrated by a single injection of benzene, phenol, or benzoquinone have been analyzed by nuclease P1-mediated 32P-postlabelling. The results showed that the RAL of DNA adduct were 9. 8-89. 13 (benzene), 20. 9243. 8 (phenol), 53. 1208. 9 (benzoquinone) adduct/108 normal nucleate. The relationship between extent of DNA adduct formation and genotoxicity of chemicals was benzoquinone>phenol> benzene.
