  • 摘要:经过镉、铅45h污染处理,运用Northein斑点杂交分析和酶活性测定的方法探测萌发期小麦ADH酶基因的表达情况。实验结果表明,ADH基因表达在RNA和酶活性两个层次均有变化,并且呈现相似的变化趋势,反映重金属的影响是来源于基因转录水平。同时,ADH基因表达量的变化是一种植物对以重金属为主导因子的环境条件的应激性反应,其中镉的效应大于铅,元素间相互作用的效应与镉铅的复合方式相关。从实验结果推测镉、铅干扰ADH基因表达的效应是间接的。
  • Abstract:After 45 hour treatment of cadmium and lead, embryoes of wheat 1257 were used to detect ADH gene expression in virtue of Northern Dot Blot and enzymatic assay. The result showed that both transcriptional quantity and enzymatic activity occurred similar alterations, which suggested that the first response was the level of transcription, and consequently passed down to the level of enzyme. Toxic effects of cadmium and/or lead were determined by the sort of heavy metal ions, their doses and the combined manner. The indirect interference of heavy metals to ADH expression of plants is proposed.
