- 摘要:真养产碱杆菌突变株B510(AlcaligeneseutrophusmutantB510)能以葡萄糖为碳源在无机合成培养基上积累聚β 羟基丁酸。为消除发酵过程中底物的抑制和优化DO stat分批补料培养中的补料速率,建立了真养产碱菌突变株B510的比生长速率μ和底物浓度S的数学模型,并求得其生长得率Yx/s.在此基础上,借助一个计算路线优化出在生长期每一个DO信号所要补加的底物量为13g/L;按中心组合实验设计优化PHB积累期最佳葡萄糖浓度为35g/L,硫酸铵为1.6g/L.通过DO-stat补料分批培养,最终细胞浓度为80g/L,PHB的产量为62g/L.
- Abstract:Glucose can be utilized by A. eutrophus mutant B510 to accumulate poly β hydrobutyric acid (PHB) in mineral medium. In order to eliminate the substrate inhibition and obtain an optimum substrate feed rate in the fed batch culture with B510,a mathematic model reflecting the relationship between the specific growth rate and substrate concentration was built and the growth yield was calculated. Based on these results, we got the optimum feed rate (13 g/L signal) for cell growth with a calculative procedure, and the optimum substrate concentration (35 g/L of glucose and 1.6 g/L of (NH4)2SO4) for PHB accumulation with centureal compsite design. The final cell biomass and PHB production in DO stat fed batch culture were 80 g/L and 62 g/L, respectively.
