- 摘要:从处理石化厂废水的活性污泥中分离出一株邻苯二甲酸酯降解菌FS1(PseudomonasfluorescensFS1),测定了P.fluorescensFS1的邻苯二甲酸酯降解酶对邻苯二甲酸二(2乙基己基)酯(DEHP)的降解特性.P.fluorescensFS1细胞中的颗粒部分、溶液部分对DEHP都具有降解作用,初步认为P.fluorescensFS1降解酶属胞内酶.P.fluorescensFS1降解酶对DEHP的最适酸度为pH6.5-8.0,温度为25-35℃,米氏常数Km为190.48nmol/mL.在好氧条件下,DEHP与P.fluorescensFS1降解酯酶作用,形成邻苯二甲酸单酯和邻苯二甲酸后,可进一步降解成苯甲酸、对羟基苯甲酸,最终转化为CO2和H2O.
- Abstract:Degradative characteristics of di(22ethylhexyl)phthalate were determined by the crude enzyme extracted from Pseudomonas fluorescens FS1 which was isolateed from the activated sludge at a petrochemical factory. The results showed that the PAEs-degrading enzyme, mainly locatized in soluble part of cytoplasm, is an intracllular enzyme. The optium temperature and pH of the PAEs-degrading enzyme activity to di(22ethylhexyl)phthalate are 20-35℃ and 6.5-8.0, respectively. The Michaelis mentn's constant (K m) of di(22ethyl hexyl)phthalate enzymatic degradation are about 190.48nmol/mL at 30℃. The enzymatic metabolism of di(22ethyl hexyl)) phthalate under aerobic conditions begins by PAEs degrading enzyme to monoester, further enzymatic degradation of monoester proceeds via phthalic acid by benzoate, 4 hydroxy benzoic acid, and then to CO2 and H2O.
