- 摘要:通过正交实验法对影响白腐菌固态发酵降解稻草木质素的5种单因素培养条件进行了优化.实验结果表明,5种因素的最佳综合水平为:菌种驯化培养8d后以0.8%(质量比)的接种量接种入发酵瓶中,于37℃培养,发酵基质的含水率为85%,同时添加0.3%的吐温80.此外,还对未经降解处理的稻草与在上述最佳发酵条件下经白腐菌降解后的稻草分别作了紫外光谱和红外光谱分析,结果表明经该菌降解后,稻草中的木质素结构被严重破坏,难降解的大分子长键烃被切断成易降解的小分子短键烃.
- Abstract:The effects of five single culture factors on lignin biodegradation ability of white-rot fungi in state-solid medium were optimized through the orthogonal experiments. The results indicated that the optimal fermentation for white-rot fungi could be reached when white-rot fungi was cultured after 8 days, 0.8% white-rot fungi (weight ratio) was inoculated, temperature was kept at 37℃, water content of the solid substrate was maintained by 85% and 0.3% Tween 80 was added respectively. In addition, the structure of the untreated straw and the treated straw degraded under the optimal fermentation condition was analyzed through ultraviolet spectroscopy and infrared spectroscopy. It could be observed that long-bond hydrocarbons of giant molecule difficult to be biodegraded were degraded into short-bond hydrocarbons of small molecule, which could be easily biodegraded. Compared with the structure of untreated straw, the structure of the treated straw was greatly altered.
