  • 摘要:利用镰刀菌(Fusarium sp.)HJ01降解中性艳蓝GL,经扩增翻译延伸因子TEF1对HJ01菌株做了种的鉴定,并考察了温度、pH值对该菌株降解酶漆酶的活性,以及对中性艳蓝GL脱色率及COD去除率的影响.结果表明,该菌株属层出镰刀菌(Fusarium proliferatum).HJ01菌株所产降解酶漆酶在pH为2.0~8.0内相对活性在12%~96%之间,在pH=4.0时,相对活性最大;在25~65℃内,随着温度的升高,漆酶相对活性增大;在65~85℃内,随着温度增大,漆酶相对活性降低.HJ01菌株在25℃、50r·min-1条件下,于液体培养基中培养96h后加入到25mL含100mg·L-1中性艳蓝GL的溶液中,在35℃、50r·min-1、pH=4.0条件下继续培养100h后,中性艳蓝GL的脱色率达100%,培养至120h后,COD去除率达87.8%.紫外光谱分析表明,中性艳蓝GL的发色基团蒽醌环被漆酶打破.
  • Abstract:The study on neutral brilliant blue GLdecoloration and degradation characteristics was carried out by the Fusarium sp. HJ01 strain. The HJ01 was identified as Fusarium proliferatum through TEF1(translation elongation factor 1 alpha). The effects of temperature and pHvalue on neutral brilliant blue GLdecoloration rate and COD removal rate were investigated. The results showed that the relative activity of laccase was between 12% and 96% in pHrange from 2.0 to 8.0, and reached its peak at pH=4.0. The relative activity increased with temperature increasing in the range from 25℃ to 65℃, but it was opposite when from 65 ℃ to 85 ℃. The HJ01 mycelia incubated in the liquid culture for 96h at 25℃ and 50 r·min-1 were added to the neutral brilliant blue GLsolution(100mg·L-1, 25mL), and were cultured continuously for another 100h at 35℃、50 r·min-1 and pH=4.0, the decoloration rate of neutral brilliant blue GLwas up to 100%, after 120h, CODremoval rate was up to 87.8%. The ultraviolet-visible spectrum showed that the chromophore anthraquinone ring of neutral brilliant blue GLwas broken down by laccase.
