  • 摘要:从堆肥中筛选到一株能在55℃生长并分泌高温α-淀粉酶的菌株,经初步鉴定为地衣芽孢杆菌,命名为Bacillus licheniformisZD-8.在液体筛选培养基的碳源为乳糖、氮源为(NH4)2SO4,通气量为10%、pH值及C/N比分别为8和3/1、65℃的条件下,培养24h的菌株产酶量最高.酶学性质研究表明,该酶最适pH值为8,最适反应温度为95℃;95℃下保温60min后酶活仅损失4.8%;较低浓度Ca2+存在下,该酶即可有很好的稳定性.污泥有机质降解实验表明,培养了22h的菌株对污泥有机质降解能力最强;将高温α-淀粉酶活性增加2倍后(为2×104U·mL-1),添加到经过不同处理的污泥中,其有机质降解率、脱氢酶增加率和SOUR增加率平均增加17.1%、76.9%、27.3%;在污泥中添加0.1mmol·L-1CaCl2后,其有机质降解率、脱氢酶增加率、SOUR增加率平均增加7.1%、20.4%、3.4%.
  • Abstract:Athermo-α-amylase producing strain was isolated from sewage municipal sludge compost and primarily identified as a kind of thermophilic Bacillus licheniformis ZD-8. As lactose and (NH4)2SO4 were employed its carbon and nitrogen sources, respectively, its maximum enzyme production of the strain was observed at 24h for the incubation conditions of medium pHof 8, ventilation of 10%, fermentation temperature of 65℃ and C/Nratio of 3/1. The optimum reaction conditions of the enzyme were determined as 95℃ and pH 8.0. The further experiments showed that the enzyme was considerable stability for only 4.8% of its activity was lost when treated at 95℃ for 60min, moreover, it became more stable when low concentration of Ca2+ existed. The experiments of sludge organic matter degradation showed that the strain incubated for 22h possessed an optimal degradation activity. Further more, the results showed that the removal rate of organic matter, increase rate of dehydrogenase and increase rate of SOURfor double activity enzyme were 17.1%, 76.9%, 27.3% higher than those of the treatments with single activity enzyme. At the same time, the addition of 0.1mmol·L-1 CaCl2 lead to their increases of 7.1%, 20.4%, 3.4%, respectively.
