  • 摘要:观察不同剂量的邻苯二甲酸二丁酯(DBP)对成年大鼠生精细胞损伤、端粒酶和端粒酶逆转录酶(TERT)表达影响.对40只健康雄性SD大鼠随机分成4组,分别为500mg·kg-1·d-1、250mg·kg-1·d-1、50mg·kg-1·d-13个染毒组和1个对照组,灌胃量为5mL·kg-1体重,灌胃法连续给药6周.对体重、脏器重等指标进行称量,对附睾中精子密度、精子畸形率等进行检测,免疫组化法检测大鼠生精细胞端粒酶活性的表达,核酸原位杂交检测睾丸生精细胞TERT的表达.结果表明与对照组比较,中、高剂量组体重、睾丸、附睾及睾丸脏器系数指标有统计学意义(p<0.05);体重、右侧睾丸和右侧附睾与染毒剂量呈负性相关(r=-0.7710,p<0.001;r=-0.6389,p<0.001;r=-0.6185,p<0.05).高、中剂量组精子密度、精子活率与对照组和低剂量组比较均降低,差异均有统计学意义(p<0.05);高、中剂量组总畸形率分别与对照组和低剂量组比较均增高,差异均有统计学意义(p<0.05).对照组、低剂量组、中剂量组及高剂量组生精小管TERT表达明显下降,染毒组生精小管TERT表达数分别低于对照组,数目也明显减少,差异均有统计学意义(p<0.05).研究表明:一定剂量的邻苯二甲酸二丁酯(DBP)可通过抑制生精细胞TERT的表达,使端粒酶活性降低从而导致生精细胞损伤,对大鼠的生长发育及睾丸功能具有毒性作用.
  • Abstract:We observed the apoptosis,expression of telomerase and telomerase reverse transcriptase (TERT) activity in spermatogenic cells after administration of different doses of dibutylphthalate (DBP) in the testes of adult male rats.Forty healthy male Sprague-Dawley rats were divided randomly into four groups and were given 0 (control group),50,250 and 500 mg·kg-1 DBP in aqueous solution by intragastric administration for six weeks.The amount of intragastric administration was 5 mL·kg-1. The body and viscera weights were measured,and sperm density and rate of abnormality in the epididymis were detected.The telomerase activity and TERTexpression in rat testicle spermatogenic cells were tested by means of immunohistochemistry (ICH) and in situ nucleic acid hybridization (ISH).Compared with that in the control group (p<0.05),the body weights,testes,epididymis and the coefficient of testicular viscera of the intermediate dose group and the high dose group have statistically significant differences.Anegative correlation was obvious among body weight,right testis,right epididymis and the dosage of DBPin water solution (r=-0.7710,p<0.001;r=-0.6389,p<0.001;r=-0.6185,p<0.05).The densities and motility rates of sperm in intermediate and high dose groups also decreased significantly over that of control and low dose groups (p<0.05).The malformation rates in intermediate and high dose groups also increased significantly over that of control and low dose groups (p<0.05).The expression of TERTof seminiferous tubules in the intermediate dose group and the high dose group all decreased significantly compared with that in the control group (p<0.05).The research shows that one of the mechanisms of male reproduction toxicity of DBP might be decrease of the telomerase activity and damage of spermatogenic cells,by means of inhibiting the expression of TERT.DBP intake has toxic effects on mammalian development and testis functions.
