  • 摘要:从稳定运行处理NO废气的生物转鼓过滤器中筛选分离到1株好氧反硝化细菌DN3,发现该菌在好氧条件下能有效去除培养液中的硝酸根,培养28h后的反硝化率大于80%.扫描电镜结果显示,菌株DN3为短杆菌,带有鞭毛,大小为(0.4~0.8)μm×(2.0~2.5)μm,菌落表面呈现乳白色.通过生理生化特性及16SrRNA基因序列研究分析,初步判断菌株DN3为假单胞菌属中的Pseudomonas putida.菌株DN3的好氧反硝化性能测试结果表明,该菌的对数生长期较长,在pH为7.5时反硝化能力最强,而在pH低于5的酸性环境中无法存活.菌株DN3对浓度低于400mg·L-1的硝酸根离子具有较好的处理效果.碳源对DN3的反硝化作用影响明显,且葡萄糖和琥珀酸钠的利用率优于甲醇、甘油和碳酸氢钠.
  • Abstract:An aerobic denitrifying bacterium(named DN3) was isolated from a NO-fed rotating drum biofilter using selective medium.The denitrifying rate of strain DN3 was more than 80% under aerobic conditions after 28 hours.The morphology of strain DN3 was a brevibacillus with an average size of(0.4~0.8) μm×(2.0~2.5) μm and an ivory-white surface.It was identified as Pseudomonas putida based on its biochemical/morphological characteristics and physiological analysis of its 16SrRNAsequence.Strain DN3 had a long logarithmic growth phase and the optimum pHwas 7.5.It could not survive at a pHbelow 5.0.It could effectively biodegrade nitrate solutions with concentrations below 400 mg·L-1.The carbon source was an important factor for the growth of DN3,and the best carbon sources were glucose and monosodium succinate.
