- 摘要:从长期受染料污染的土壤中分离出一株能对甲基橙高效降解脱色的柠檬酸盐杆菌Citrobacter sp. LW-3.菌株LW-3 在添加了0.5%(g/100 mL)葡萄糖的MSM-1培养基中,16 h使100 mg·L-1甲基橙降解掉90.56%.该菌株对甲基橙降解脱色可在完全好氧条件下,脱色与木质素过氧化物酶、NADH-DCIP原酶、核黄素还原酶有关.菌株 LW-3 降解甲基橙的适宜温度较广,20~40 ℃ 24 h均能使超过 80% 的甲基橙降解脱色,适宜pH范围为6.0~8.0.通气量对菌株 LW-3 影响较小,24 h各装液量组(25~200 mL)甲基橙均能降解近90%.同时,菌株 LW-3具有较广的染料降解谱,3 d 内可使 20 mg·L-1另3种偶氮染料、两种三苯甲烷类染料和一种蒽醌类染料很好脱色.菌株 LW-3 在染料废水的生物处理方面有较强的实际应用价值.
- Abstract:The strain Citrobacter sp. LW-3, capable of efficiently degrading and decolorizing methyl orange, was isolated from the soil contaminated by dyes for many years. It was found that in medium MSM-1 containing 0.5% (w/v) gluose, the strain LW-3 could degrade 90.56% of 100 mg·L-1 methyl orange within 16 hours. The whole process of strain LW-3 degrading and decolorizing methyl orange could be under aerobic condition, and the decolorization might have the relationship with lignin peroxidase, NADH-DCIP reductase and riboflavin reductase enzyme. The favorable temperature range for LW-3 removing methyl orange was wide. In the range of 25 to 30 ℃, the strain LW-3 could decolorize more than 80% of methyl orange in 24 h. The favorable pH range was 6.0~8.0 for the strain degrading methyl orange. Changes of ventilation condition had a little influence on the degradation, and in all liquid volume group (from 25 to 200 mL), nearly 90% of methyl orange were decolorized in 24 h. At the same time, the strain LW-3 had a broad degradation spectrum for dyes, and it could effectively decolorize 20 mg·L-1 of the other three kinds of azo dyes, two kinds of triphenylmethane dyes and a sort of anthraquinone dye in 3 days. The results suggested that applying the strain LW-3 in treating wastewaters containing various kinds of dyes was feasible in practical situations.
