堆肥过程中通气方式对碳氮代谢相关基因表达影响的研究

- 摘要:采用实时荧光定量PCR对两种不同强制通气方式(罐体内嵌式通气和整体式通气)好氧发酵过程中碳氮代谢相关基因表达,包括细菌16S rDNA、β-glucosidase、amoA、nxrB1基因进行了定量分析,并对比研究了发酵过程中理化参数及堆体中碳氮代谢相关基因表达的变化.实验结果表明,在堆肥过程中,罐体内嵌式通气发酵堆体细菌总量大于整体式通气发酵;罐体内嵌式通气与整体式通气发酵中β-glucosidase基因拷贝数分别在堆肥第7 d与第4 d达到最大值,该数值分别是堆肥第1 d的4.0倍和6.2倍,表明发生了强烈的生物降解作用,碳代谢活动剧烈;氨氧化细菌(AOB)在强制通气好氧发酵的高温期含量极少,直至腐熟期才大量积累,导致大量铵态氮转化为氨气造成氮损失;高温对nxrB1基因表达有严重阻碍作用,在腐熟阶段罐体内嵌式通气方式发酵中nxrB1基因优先于整体式通气发酵表达并达到峰值.
- Abstract:Quantitative analysis of genes related to carbon and nitrogen metabolism, including bacterial 16S rDNA, β-glucosidase, amoA, and nxrB1, was investigated by real-time PCR in two different aeration (built-in aeration and overall aeration) composting processes. The experimental results showed that total bacteria in built-in aeration mode was more than that of overall aeration mode; β-glucosidase gene copies in built-in aeration and overall aeration mode reached the maximum value on day 7 and day 4, respectively, which was 4.0 and 6.2 times that on day 1, indicating strong biodegradation and carbon metabolic activity during composting process. AOB content was low in thermophilic stage and high in maturation, which may lead to a large number of ammonia nitrogen lost during this stage. High temperature had an opposite effect on the expression of nxrB1 gene, and the gene expression in built-in aeration was more than overall aeration during the maturation period.
