  • 摘要:采用海藻酸钠包埋和戊二醛交联法固定碳酸酐酶,比较游离酶和固定化酶的酶学性质,并用固定化酶催化吸收CO2.实验结果表明:固定化酶的酶活力回收率是56.3%;最适反应温度为30 ℃,最适pH为7,在T=30~40 ℃和pH=6~10区间有稳定和很高的相对酶活力;固定化酶在60 ℃温浴1 h后,能保持39.8%的相对酶活力,而游离酶已经基本完全失去活性,固定化酶在pH=5.0~11.0范围内pH稳定性显著;固定化酶在T=30 ℃下催化吸收CO2效果显著,10%的反应时间完成了80.2%的催化反应,催化吸收CO2的量是空白试验的4.2×103倍,SEM显示反应后的固定化酶表面出现大量形状不规则的小孔,空隙结构变得发达;固定化酶具有良好的储存稳定性和操作稳定性.
  • Abstract:The carbonic anhydrase (CA) was embedded with sodium alginate, which was cross-linked with glutaraldehyde. The enzymatic properties of CA were compared between free and immobilized enzyme, and CO2 catalytic absorption with immobilized CA was studied. The results show that the immobilized CA retains 56.3% of its original activity. The opti-mum pH of immobilized CA is 7, and temperature is 30 ℃. The immobilized CA has high stability and activity at the range of 30~40 ℃ or pH=6~10. The free enzyme lost all its activity when heated at 60 ℃ for 1 h but the immobilized enzyme kept its activity at 39.8%. The pH value of immobilized CA was stable at the range of pH=5.0~11.0. Immobilized enzyme shows significant effect on CO2 catalytic absorption at 30 ℃. The 80.2% of catalytic reaction took 10% of the reaction time, and catalytic absorption amount of CO2 is 4.2×103 times compared to that of the control experiment. The surface of immobilized CA appeared a large number of irregular-shaped holes, and the pore structure becomes rich through scanning electron microscopy (SEM) observation. Furthermore, the immobilized CA enhances enzamatic stability and durability.
