  • 摘要:研究了温度、转速、pH、接种量、碳源和亚硝酸盐氮浓度对耐冷反硝化细菌Pseudomonas putida Y-12去除亚硝酸盐氮和总氮的影响,在此基础上,初步考察了反硝化脱氮产物中温室气体(CO2和N2O)的含量,探索了对富营养化景观水体中亚硝酸盐氮和总氮的去除能力.结果表明:Y-12菌株去除亚硝酸盐氮和总氮的最佳碳源为柠檬酸钠,在15℃和pH为7.2条件下,该菌以柠檬酸钠为唯一碳源对亚硝酸盐氮和总氮去除率分别达到了100%和81.8%; 反硝化脱氮产生了19559.82 mg·m-3 CO2和0.11 mg·m-3 N2O;在15℃,150 r·min-1摇床培养条件下,48 h内菌株Y-12对景观水体中亚硝酸盐氮和总氮去除率分别达到了37.7%和51.5%,表明该菌在初春及晚秋水体的低温时段具有较好的应用潜力.
  • Abstract:A nitrite-denitrifying bacteria named Y-12 was isolated from the long-term flooded paddy soil at 10℃, which exhibited high efficient aerobic denitrification ability with nitrite as sole nitrogen source. Conditions, including initial pH, shaking speed, carbon source, temperature, inoculated quantity and initial nitrite content of medium were studied and selected based on the nitrite and total nitrogen removal efficiencies. Meanwhile, N2O and CO2 emission flux were measured during denitrifying process and the abilities of nitrite and total nitrogen removal rates were analyzed preliminarily in the eutrophic landscape water. The optimal incubation conditions for the aerobic nitrite-denitrifying strain Y-12 were:pH, 7.0~8.0; inoculums size, 2.0×106 CFU·mL-3; temperature, 20℃; carbon, sodium citrate; shaking speed, 200 r·min-1; nitrite concentration, 10 mg·L-1. Results showed that the strain Y-12 could remove nitrite and total nitrogen up to 100% and 81.8% respectively, with sodium citrate as sole carbon source at 15℃ in 48 h. High oxygen and low nitrite concentrations were beneficial for strain Y-12 in denitrification. Flux of CO2 was 19559.82mg·m-3 and it was 0.11 mg·m-3 for N2O by strain Y-12 during denitrifying. In addition, the nitrite and total nitrogen removal efficiency were up to 37.7% and 51.5% respectively, within 48h, at 15℃, 150 r·min-1 to dispose the eutrophic landscape water. Results indicated that the strain Y-12 is a potential strain in N removing in the early spring and late autumn.
