  • 摘要:研究小鼠腹腔巨噬细胞(PMФ)对于环境污染物镉、铅的细胞毒性反应,探讨巨噬细胞作为生物监测指示物的意义.实验取小鼠腹腔巨噬细胞体外培养,分别经不同浓度氯化镉、醋酸铅染毒后,用电镜观察细胞形态变化,用酶标仪检测其还原MTT能力、吞噬功能及NO产量的变化.结果显示,氯化镉、醋酸铅染毒后(10-4 mol·L-1),小鼠腹腔巨噬细胞突起消失,细胞死亡;巨噬细胞还原MTT能力显著下降(p < 0.05),NO合成明显减少(p < 0.05),且具有明显的剂量-效应关系.实验结果说明,氯化镉、醋酸铅对小鼠腹腔巨噬细胞具有明显的细胞毒性.结果提示,PMФ可作为生物监测指示物,应用于环境污染的生物监测.
  • Abstract:To study the toxicity effects of environmental pollutant cadmium chloride or lead acetate on the peritoneal macrophages (PMФ) of mice, and to evaluate the potential application of macrophages as a biomarker in biological monitoring, PMФ were exposed to variable concentrations of cadmium chloride or lead acetate in vitro. After exposed to Cd or Pb, morphologic changes of PMФ were detected by using the electron microscopy and the PMФ activity were examined by measuring cell viability, phagocytosis activity and nitric oxide production. Marked morphologic changes of PMФ were observed after exposing to 10-4 mol·L-1 cadmium chloride or lead acetate. MTT deoxidizing ability and NO production were obviously inhibited (p<0.05) in a dose-dependent manner. Results show that PMФ were damaged similarly by using cadmium chloride or lead acetate, indicating PMФ could be used as an effective biomarker for biological monitoring for heavy metal pollutants.
