  • 摘要:碳源对微生物异养硝化至关重要,为了探究碳源对耐低温异养硝化菌-哈尔滨不动杆菌HITLi 7T低温生长和氨氮代谢的影响,考察了乙酸钠、碳酸钠、淀粉、柠檬酸钠、蔗糖、葡萄糖、甘油、糊精、乳糖和麦芽糖10种碳源.结果表明,碳源种类对HITLi 7T低温生长速率和胞外分泌物含量影响较大.乙酸钠是HITLi 7T的最优碳源,低温下培养4 d后OD600能提高到0.146,产生的ATP也明显高于其他组,为5.124 μmol·mg-1·prot-1;此时,总胞外分泌物量最多,为3.86 mg·L-1,多糖占比也最高为41.7%;4 d氨氮平均降解速率最大,为0.042 mg·L-1·h-1;其次为葡萄糖和碳酸钠.同时考察了不同碳氮比的影响,结果表明HITLi 7T以乙酸钠为碳源时,最佳C/N比为8,此时氨氮去除速率最高为0.216 mg ·L-1 ·h-1,总EPS量为7.04 mg ·L-1,其中多糖占比为55.3%.
  • Abstract:In order to investigate the effect of carbon sources on the ammonia removal and the growth of heterotrophic nitrifier, Acinetobacter harbinensis HITLi 7T, at low temperature, sodium acetate, sodium carbonate, starch, sodium citrate, sucrose, glucose, glycerol, dextrin and lactose were tested. Results found that the type of carbon source had a great influence on the growth and EPS secretion of HITLi 7T at low temperature. Sodium acetate was the optimal carbon source and when served as the sole, after 4-day cultivation, the OD600 of HITLi 7T reached 0.146, with the ATP at 5.124 μmol·mg-1·prot-1 and the average ammonia removal rate achieved 0.042 mg·L-1·h-1. The performance of Glucose and sodium carbonate are next to sodium acetate. In addition, different type of carbon sources also had significant effects on extracellular polymeric substances (EPS) secretion of HITLi 7T. When sodium acetate was the carbon source, the EPS amount of HITLi7T was 3.86 mg·L-1, with 41.7% of polysaccharide.It could be found that low C/N ratio was more suitable for HITLi 7T. Ammonia removal rate reached maximum at 0.216 mg·L-1·h-1 as C/N was 8. At the same time, the EPS amount was 7.04 mg·L-1 with 55.3% polysaccharide.
