  • Abstract:Index of biotic integrity (IBI) has been widely applied in health assessment of rivers. However, with the increasing anthropic impact on urban rivers, the biodiversity losses of local macroorganism species reduced the accuracy of bioassessment. Microbial communities thus have been proved to be more sensitive indicators of ecological health in highly impacted aquatic environment. This study applied illumina high-throughput DNA sequencing technique to analyze the microbial community diversity and composition of the selected 8 urban rivers in Wuxi city in winter 2018 and spring 2019. A dataset of candidate metrics was first compiled, and classified into three categories:composition/abundance, richness/diversity, and sensitivity/tolerance. This study improved the traditional candidate indicator screening method, conducted difference analysis, boxplot test, and Speraman correlation, and ultimately developed the microbial biological integrity index (M-IBI) framework for habitat evaluation in winter and spring. The results showed that M-IBI was able to effectively discriminate different impaired site groups, and had a good correlation with the index of water quality and the physical habitat index, indicating that the M-IBI is a reliable and novel assessment tool for assessing the ecological health status of urban rivers.
